Nitric Oxide is detected in biological fluids via nitrite. Additionally, nitrate can be reduced to nitrite by NADPH in the presence of the enzyme Nitrate Reductase (NR). The nitrite formed reacts with sulfanilamide and N-(1-naphthyl)-ethylenediamine dihydrochloride instantaneously to give a red-violet diazo dye. The diazo dye is measured by its absorbance at 540 nm.
